The ACA ELISA is intended for use in the detection of Anticardiolipin Antibodies in human serum or plasma. ACA can be used as the indicators of anti-phospholipid antibody syndrome (including the formation of thrombosis), spontaneous abortion, thrombocytopenia and the diagnosis of CNS lesions. ACA-positive IgG-type may indicate that patients with cerebral thrombosis; it also result in spontaneous abortion. The incidence of thrombocytopenia in Positive ACA patient is significantly higher than negative, and is related to the degree of platelet reduction, and ACA-positive is significantly associated to SLE patients with central nervous system (CNS) thrombosis.
Summary and Explanation
Purified cardiolipin antigen is coated on the surface of microwells. Patient serum is added to the wells, and the ACA, if present, binds to the antigen during incubation. After washing the wells to remove unbounded sample, antibody to human conjugated with HRP is added. Unbounded conjugate is removed by a subsequent washing step. A solution of TMB Reagent is then added to the microwells. The enzyme conjugate catalytic reaction is stopped at a specific time. The intensity of the color generated is proportional to the amount of ACA in the sample.
