The ANA-antibody ELISA is intended for use in the detection of ANA antibody to the virus in human serum. Systemic autoimmune diseases are characterized by the presence of antinuclear autoantibodies (ANA).Higher titers of ANA is highly indicative of lupus erythematosus, though they also appear in some other auto-immune diseases such as drugs –induced lupus erythematosus, overlap syndrome,mixed connective tissue disorder, scleroderma, polymyositis & dermatomyositis ,Sjögren's syndrome, rheumatoid arthritis, autoimmune hepatitis, Hashimoto's and myasthenia gravis.
Summary and Explanation
Purified ANA antigen is coated on the surface of microwells. Patient serum is added to the wells, and the ANA antibody, if present, binds to the antigen during incubation. After washing the wells to remove unbound sample, antibody to human conjugated with HRP is added. Unbounded conjugate is removed by a subsequent washing step. A solution of TMB Reagent is then added to the microwells. The enzyme conjugate catalytic reaction is stopped at a specific time. The intensity of the color generated is proportional to the amount of ANA antibody in the sample.
