The ASA ELISA is intended for use in the detection of Anti-Spermatozoa Antibodies in human serum. The rate of ASA positive is usually between 10-30% in infertility patients, serum, especially obstructive azoospermia patients, ASA positive rate can be as high as 60%, high ASA titer is the vital reason that can result in autoimmune infertility. The ASA detection offers a valuable prognostic indicator for infertility treatment.
Summary and Explanation
Purified spermatozoa antigen is coated on the surface of microwells. Patient serum is added to the wells, and the ASA, if present, binds to the antigen during incubation. After washing the wells to remove unbounded sample, antibody to human conjugated with HRP is added. Unbounded conjugate is removed by a subsequent washing step. A solution of TMB Reagent is then added to the microwells. The enzyme conjugate catalytic reaction is stopped at a specific time. The intensity of the color generated is proportional to the amount of ASA in the sample.
